Adamax is a nine-amino-acid synthetic peptide derived from Semax, itself an analog of the ACTH(4-10) fragment originally developed at the Institute of Molecular Genetics of the Russian Academy of Sciences. Adamax modifies the Semax sequence in two ways: an acetyl group on the N-terminus (forming the more stable N-acetyl-Semax intermediate) and a bulky, lipophilic adamantane diamondoid group on the C-terminus (the structural motif characteristic of Peptide 021).


These two modifications are the basis of its differentiated profile. The N-acetyl group resists premature enzymatic degradation in circulation and improves passage across the blood-brain barrier, while the adamantane group sterically shields the C-terminus from proteolytic cleavage, substantially extending the molecule's functional half-life relative to native Semax — from a window of hours to roughly a full day per dose. The net effect is a longer, more stable arc of neuromodulatory activity.


Mechanistically, Adamax is associated with upregulation of brain-derived neurotrophic factor (BDNF) and enhanced sensitivity of hippocampal TrkB receptors, alongside modulation of dopaminergic and serotonergic signaling and neuroprotective pathways. BDNF supports the development, maintenance, and protection of neurons, processes tied to learning, memory consolidation, and overall neural plasticity. Adamax is considered among the most potent of the Semax-derivative class.
